Mechanical properties and drug loading rate of a polycaprolactone 5-fluorouracil controlled drug delivery system

Abstract In the context of precision medicine, controlled drug delivery systems (CDDSs) have become a research focus. The structural integrity CDDSs is critical for ensuring an acceptable release rule; hence, CDDS must possess appropriate mechanical properties. this study, polycaprolactone/5-fluorouracil (PCL/5-Fu) was fabricated via solvent evaporation, and effects PCL molecular weight 5-Fu loading rate on properties were evaluated. results tensile testing, scanning electron microscopy, substance analysis indicated that when content less than 9.09% in developed CDDS, completely compatible with PCL, no crystal aggregation induced. addition, maximum required to retain 23.08%, corresponding strength sample 12.9 MPa. This sufficient prevent failure instantaneous due reduction during application system. In-vitro experimental demonstrated PCL/5-Fu can achieve over 1000 h. These findings provide basis establishing model proposed CDDS.
